Screening of drug concentration of resveratrol in POI rats

Abstract:

Objective To observe the curative effect of different concentrations of resveratrol on rats with premature ovarian insufficiency (POI) and to determine the optimal drug concentration of resveratrol for treating premature ovarian insufficiency dysfunction in rats. Methods A total of 90 5-week-old SPF grade SD healthy female rats were randomly divided into a low-dose resveratrol group (18 rats in group a) and a middle-dose resveratrol group (18 rats in group b), a high dose resveratrol group (18 rats in group c), a model group (18 rats in group d) and a blank control group (18 rats in group e). Group a, group b, group c were administered with a daily dose of 20 mg/kg, 50 mg/kg, 80 mg/kg by gavage, respectively; the model control group was administered with 0.5% sodium carboxymethyl cellulose solution by gavage. ELISA was used to detect the level of serum sex hormone levels: anti-Mullerian hormone (AMH), luteinizing hormone (LH), follicle stimulating hormone (FSH), estradiol (E2) level and HE staining of rats to observe unit ovarian area of mature follicles, atresia follicles and corpus luteum, evaluation of resveratrol efficacy. Results Rats treated with different concentration of resveratrol had a significantly increased level of serum AMH and E2 compared to rats treated with 0.5% carboxymethyl cellulose solution (P<0.05), LH and FSH levels were lower (P<0.05).The serum level of AMH and E2 in POI rats treated with resveratrol at different concentration were lower (P<0.05) and LH and FSH were higher (P<0.05) than those of rats in the blank control. Compared with rats in group a, b, c and e, the number of granulosa cells, mature follicles and corpus luteum in ovaries was smaller in group d (P<0.05); In contrast, the number of atresia follicles was significantly increased in group d(P<0.05).  Conclusion Intragastric treatment with different concentration of resveratrol of all have a certain effect on POI rats receiving, with 50 mg/kg showing the best result, and thus being the best drug concentration.
