Comparative study on the production of loose connective tissue tear in rats

Abstract:

Objective To compare the difference of loose connective tissues from the subcutaneous and mesenteric membrane of Wistar rats. To compare the difference in staining of loose connective tissue from different sites and with different staining methods.  Methods Wistar rats were continuously injected intraperitoneally with 2.5 ml of trypan blue saline solution (10 g/L) once a day for three days. Loose connective tissues were taken from the subcutaneous and mesentery of rats, respectively. Two sites were treated with HE staining and aldehyde fuchsin-bright green-orange yellow G staining. Results In subcutaneous extraction with HE staining, a large number of fibroblasts could be seen. Mast cells were obvious and macrophages were visible. Elastic fibers and collagen fibers were visible, but not obvious. In Aldehyde fuchsin-bright green-orange G staining, elastic fibers and collagen fibers were stained into blue-violet and collagen Orange, respectively. But cells were not colored. In mesenchymal material with HE staining, fibroblasts, mast cells and macrophages could be found. Elastic fibers were stained into blue and purple, while collagen fibers were stained into light red. Aldehyde fuchsin-bright green-orange G staining stains elastic fibers and collagen fibers into red and Bright green, respectively. The color of mast cells was red, and the nucleus was brown. Macrophages were clearly visible with irregular shape. Thick blue-violet particles could be seen in the cytoplasm, and the nucleuses are round and brown. The cytoplasm of fibroblast cytoplasm could be colored, while the nucleuses were stained into brown. Conclusion Aldehyde fuchsin-bright green-orange G staining for loose connective tissue from inferior mesenteric is a simple and suitable method for stretch preparation of loose connective tissue in rats.
